چکیده

Background: Ursodeoxycholic acid is an important clinical drug in the treatment of liver disease. In our previous work, ursodeoxycholic acid was prepared by electroreduction of 7-ketolithocholic acid. The separation of ursodeoxycholic acid from the electroreduction product (47% (w/w) ursodeoxycholic acid) by silylation crystallization is described herein. Results: N,N-dimethylformamide was used as the solvent, whereas hexamethyldisilazane was the reaction agent. The optimal material ratio of electroreduction product/N,N-dimethylformamide/hexamethyldisilazane was found to be 1:10:2 (w/v/v). The reaction proceeded for 2 h at 60°C, and the corresponding silylation derivative was separated by crystallization and pure ursodeoxycholic acid was recovered by 5% acid hydrolysis at 50°C for 0.5 h. The maximum recovery and purity of ursodeoxycholic acid were 99.8% and 99.5%, respectively. Conclusion: Ursodeoxycholic acid with high purity and high recovery can be prepared directly. The developed method offers a potential application for large-scale production of ursodeoxycholic acid.

Application of ursodeoxycholic acid in hepatobiliary scintigraphy for neonatal hyperbilirubinemia: Comparison with phenobarbital

Introduction: Early differentiation of biliary atresia from neonatal hepatitis is of utmost importance, since on time surgery of biliary atresia significantly improves the outcome. Hepatobiliary scintigraphy is an integral part of diagnosis work-up of these patients; however its specificity for diagnosis of biliary atresia is suboptimal. In this study we evaluated t...

Sulfuric acid ([3-(3-silicapropyl)-sulfanyl]propyl)ester as recyclable catalyst for the silylation of hydroxyl groups

Sulfuric acid ([3-(3-silicapropyl)sulfanyl]propyl]ester (SASPSPE) is employed as a recyclable catalyst for the silylation of hydroxyl groups. A good range of primary, secondary alcohols and phenolic hydroxyl groups were effectively converted into their corresponding trimethylsilyl ethers with hexamethyldisilazane in the presence of catalytic amounts of SASPSPE at room temperature with short rea...

Design of Optimal Process Flowsheet for Fractional Crystallization Separation Process

A procedure is presented that synthesizes fractional crystallization separation processes to obtain pure solids from multi-component solutions. The method includes a procedure to generate a network flow model to identify alternative process designs for fractional crystallization. The main advantage of this systematic procedure with respect to other reported procedures is using non-equilibri...

Thin-layer chromatographic separation of conjugates of ursodeoxycholic acid from those of litho-, chenodeoxy-, deoxy-, and cholic acids.

Separation of the glycine and taurine conjugates of ursodeoxycholic acid from those of lithocholic acid, chenodeoxycholic acid, deoxycholic acid, and cholic acid by thin-layer chromatography is described. Thus, on running a silica gel G plate first in a solvent system of n-butanol-water 20:3 and then in a second solvent system of chloroform-isopropanol-acetic acid-water 30:20:4:1, all the above...
